Those are pipes for the PCV system. The one on the right is fresh air feed for the PCV system and the one on the left goes to the intake manifold, to operate the PCV valve attached to the cam cover via vacuum.
 
If you're planning to take the cam cover out for painting, then no. As long as you put them back in after you've finished.

Running the engine without the pipes would be something to avoid though.
